  Features  
  Rapid photobleaching of cell probes labelled with fluorochromes still hampered qualitative and quantitative assessment of the label.
Squarix ImmunoSelect® Antifading Mounting Medium is developed for fluorescence-microscopical use combining strong initial fluorescence with drastical reduction of photobleaching over time. In contrast to several media, Squarix ImmunoSelect® Antifading Mounting Medium does not exhibit autofluorescence at any common wavelength.
Squarix ImmunoSelect® Antifading Mounting Medium is an aqueous mountant which does not solidify directly, but remains a viscous liquid for several days on the slide. To mount cellular material on the slide one drop of medium is sufficient. After a cover slip is applied, the medium disperses over the entire section. When mounted slides are stored at 4°C in the dark, fluorescence will remain for many weeks.
 
  Squarix ImmunoSelect® Antifading Mounting Medium is available in three different formulations:  
 
  Mounting Medium (SQ-IS-12001)  
  Mounting Medium - DAPI (SQ-IS-12002) with 4',6-diamidino-2-phenylindole (DAPI) cell membrane permeable fluorescent stain that binds strongly to A-T rich regions in DNA  
  Mounting Medium - PI (SQ-IS-12003) with propidium iodide (PI) cell membrane impermeable fluorescent stain that intercalates into DNA  
 
  Compatibility with Fluorochromes  
  The Mounting Medium is tested for several fluorescence dyes:  
  Fluorescein derivatives, e.g. FITC  
  Rhodamine derivatives, e.g. TRITC, Texas Red  
  Cy3 and Cy5  
  Phycobilliproteins, e.g. PE  
  ALEXA dyes  
  DAPI  
  Hoechst 33358 and 3334 dyes  
  Propidium Iodide  
 
  ImmunoSelect® Antifading Mounting Medium with DAPI DNA-stain  
  ImmunoSelect® Antifading Mounting Medium DAPI contains ready to use DAPI DNA-stain. DAPI (4',6 diamidino-2-phenylindole) produces a blue fluorescence with excitation maximum at 365 nm and emission at about 460 nm. Therefore there is no emission overlap with all green or red emitting dyes.
DAPI is a common counterstain in immunohistochemistry. Stainings serve also as detection techniques for mycoplasma infection (see below).
 
 
  DAPI-Test for the detection of Mycoplasma-DNA in contaminated cells  
  Mycoplasma contaminations in cell cultures are no rare events. Following the literature about 5 - 85 % of all cell cultures are infected. Therefore mycoplasma contaminations represent a continuous risk for in vitro experiments, especially because early contaminations mostly are not detected, due to the fact, that considerable cellular effects are seen only in later stages. Positive cultures exhibit a characteristic image after DAPI staining. At low contamination in most cases a clear fluorescence at the cytoplasmic edge of the contaminated cells occur. At high contamination levels the cells look like sprinkled with powdered sugar. The DAPI test is a very easy executed test, but analysis requires a fluorescence microscope with UV-excitation. Adherent growing cells are cultivated on slides or coverslips, not adherent cells can be cytocentrifuged on slides. As fast and simple alternative, cells can be used directly after dropping on ImmunoSelect® Adhesion Slides (Cat. No SQ-IS-10050 and SQ-IS-10100). Cells will then be fixated in cold methanol for 5 minutes, mounted in ImmunoSelect® DAPI-medium and directly examined under the fluorescence microscope.  
 
  Storage  
  ImmunoSelect® Antifading Mounting Media should be stored at 2-6°C in the dark. The expiry at 2-6°C is 1 year.  
 
  Safety  
  Media contain 0.05 % sodium azide as preservative. DAPI and PI containing media are toxigenic and mutagenic and therefore they should be handled with care. Do not get in eyes, on skin, or on clothing. If contact should occur, immediately flush with copious amounts of water.
